I don't think it's hard, I do think the amount of combinations will be high though, about: 100x99x98x97x96= 9 billion possible hero combos, makes me feel like I overlooked something in my calculations...
It would be interesting to see what hero duo works good in a team.
For example if Axe + Kunkka has a winrate of 52% when they are on the same team.
There are 96 heroes, so there are 96*95*94*93*92 (7,334,887,680) PERMUTATIONS. Permutations are where the order matters, so (sven,wisp) is different than (wisp,sven).
Now, given a team of 5 players, there are 5*4*3*2*1 (120) ways to order them. So each unique team where order doesn't matter is counted 120 times in that 7 billion number. So divide the first by the second to get the number of different 5-man teams.

Regardless, that's an unwieldy number of teams to look at, an you'll run into sample size issues.
But there are only 4,560 two-person combinations, where each match will include 20 two-person combos, 10 for each team.
Honestly though, the site needs a way to filter or show results based on skill level first. Wisp-Tiny might be great, but if 90% of the wisp-tiny games are with low level players, then all it's really going to show you is how us average players do with wisp-tiny combo. (poorly i'm guessing, since wisp is not terribly easy)
I think one would have to be able to filter out low level games to pull meaningful hero-combo-strength out of the noise.
mattieshoes has an awesome point that is hard to argue against.
But for 2 person combos its got to be double stun in the safe lane that has the highest win rate (at least for that lane)
ie. Chaos knight and Centaur war runner with the combo of rift, chaos bolt, stomp, double edge can be devastating during the lane stages.
Or disable and high damage Shadow shaman and juggernaut can easily get a first blood (assuming that juggs can position a blade fury block in time)
I hope that's what you mean about combos. IMO it is a better way to look at this game in terms of checks and counters to heroes and likely strategies but in the end we are still talking about junky pubs right and people often don't pick appropriately lol.
The formula for a combination with no repetition is (n!)/((k!)((n-k)!)) so (98!)/((5!)((98-5)!)) = 67910864. While this is still nothing to laugh at in terms of size, it is nowhere near the absurd 98! (9.42689... × 10^153).

I think it would be interesting to see what hero composition has the highest winrate. So for example you would see:
Tidehunter
Wisp
Lifestealer
Luna
Omniknight
Winrate: 49.6%
I think this wouldn't simply be the top 5 winrate heroes but an actually good combo.
